Solentim iPSC Matrices: MatriClone™ and Clinical Manufacturing Grade Laminin

Support iPSC growth and pluripotency in both single and multi-cell formats

MatriClone and Clinical Manufacturing Grade Laminin: Accelerating Cell Therapies to Market

Induced pluripotent stem cells (iPSCs) have the potential to differentiate into any cell type in the human body, making them exciting candidates for the development of new cell therapies. However, without the support of crucial elements from the basement membrane, the pluripotent potential of these cells is at risk.

MatriClone and Clinical Manufacturing Grade Laminin are comprised of the recombinant laminin-511 E8 fragment which is instrumental to the growth and maintenance of iPSCs. Together, these matrices provide the perfect solution for both process development and manufacturing of new iPSC therapies.

Supporting the Development of iPSC Therapies

Supports Regulatory Approval

MatriClone and Clinical Manufacturing Grade Laminin are optically suited for imaging, allowing scientists to capture high resolution, whole well images required for regulatory approval.

Improved iPSC Adhesion

Both MatriClone and Clinical Manufacturing Grade Laminin are composed of the recombinant laminin-511 E8 fragment. While there are numerous isoforms of laminin available, the truncated E8 fragment of laminin-511 promotes greater adhesion compared to alternate isoforms, in addition to the full-length protein.

Ensures iPSC Pluripotent Potential

Maintaining pluripotency throughout the development of iPSC therapies can be a significant challenge. Fortunately, MatriClone and Clinical Manufacturing Grade Laminin provide the crucial support required throughout gene editing, single cell isolation, clonal expansion and characterization, expediting novel iPSC therapies.

Adapting iPSCs to MatriClone and Clinical Manufacturing Grade Laminin

The process of adapting iPSCs is very straightforward. Simply dissociate your cells following standard operating procedures at the time of passage. Seed the cells into a vessel which has either been pre-coated with MatriClone/Clinical Manufacturing Grade Laminin or add the matrix in-solution at the time of seeding.

Calculate your matrix needs with our user-friendly calculator!

Our iPSC matrices offer versatile application, allowing for either in-solution use or pre-coating of plates. In-solution involves adding the matrix to the culture media simultaneously with cell passaging, eliminating the need for pre-coating plates. The ideal coating density may vary based on factors such as cell line, application, and chosen medium. No need to fret—our convenient calculator is at your service! Simply specify your preference for in-solution or pre-coating, input your surface area and number of plates, and let the calculator determine the amount of matrix required.

Remember

When it comes to using MatriClone or Clinical Manufacturing Grade laminin in-solution, the important thing to remember is that the required volume of matrix is independent of the well fill volume, it’s all about surface area.

For example:

  • To coat a 6-well plate (9.6 cm2/well) with 0.25 μg/cm2 following the in-solution method, 4.8 μL of MatriClone is required per well.
  • To coat all 6 wells in the plate, 28.8 μL of MatriClone should be added to 12 mL of cell suspension before seeding 2 mL of cell suspension, plus MatriClone, into each well.
  • Should the scientist decide to fill each well with 1.5 mL of medium instead, the quantity of MatriClone would still be 4.8 μL per well because the surface area of the well has not changed.

MatriClone + VIPS PRO: Unlocking potential

VIPS PRO is an automated cell seeder and whole well imaging system which dispenses single cells and captures high-quality whole well images to provide assurance of clonality.

Pairing VIPS PRO’s high throughput seeding and imaging capabilities with MatriClone’s and Clinical Manufacturing Grade Laminin’s ability to support isolated iPSCs unlocks a new level of potential. The result? Reduced project costs and timelines, empowering scientists to achieve their goals more efficiently than ever before.

FAQs

MatriClone is a xeno-free, recombinant Laminin-511 E8 fragment produced in CHO-S cells which is designed to support the early growth of Induced Pluripotent Stem Cells (iPSCs). MatriClone supports iPSC growth and pluripotency in both single and multi-cell formats and and is optically suited for automated imaging.

Absolutely! Advanced Instruments are uniquely positioned to provide both research grade MatriClone, and Clinical Manufacturing Grade Laminin from the same source material, ensuring quality and consistency across the research and GMP iPSC workflows.*

*To purchase Clinical Manufacturing Grade Laminin, you must have Solentim instrumentation in your lab.

MatriClone and Clinical Manufacturing Grade Laminin must be used in combination with tissue culture treated plates.

The term ‘in-solution’ refers to adding the matrix to the culture media at the same time as the cells during cell passaging. As MatriClone and Clinical Manufacturing Grade Laminin are soluble, there is the option to add it during plating rather than pre-coating the plates.

Using MatriClone/Clinical Manufacturing Grade Laminin in-solution saves time as you no longer need to coat your plates ahead. Simply add the matrix to your cell suspension and seed directly into a tissue culture treated plate!

Additionally, in-solution use requires much less matrix compared to pre-coating.

Please note that MatriClone/Clinical Manufacturing Grade Laminin must be used in-solution when seeding with Advanced Instruments’ VIPS PRO.

Yes, however this process is very simple. Once cells have reached confluence and are ready to be passaged, simply dissociate them following your standard protocol. Instead of plating the cells onto your traditional matrix, seed your cells into a plate which has been pre-coated with MatriClone/Clinical Manufacturing Grade Laminin, or add MatriClone/Clinical Manufacturing Grade Laminin during plating (if following the in-solution protocol).

Cells can be dissociated using any of the traditional dissociation reagents. Internally, we have experience with Accutase, TrypLE, Versene, ReLeaSR, GDCR and EDTA. We have found TrypLE and ReLeaSR to be the most popular amongst our customer base.

Yes, the matrices are suitable for clump and single cell passaging of iPSCs.
