Comparison Between InstiGRO™ CHO, InstiGRO CHO PLUS

In this case study we will examine a customer’s evaluation of the ACF InstiGRO CHO and InstiGRO CHO PLUS cell culture supplements.

Like many cell line development departments out there, this customer was struggling with little to 0% cloning efficiency. The customer had a handful of problematic CHO cell lines on their hands, all of which were expressing monoclonal antibodies. After reaching out to Advanced Instruments and learning more about how InstiGRO supports clonal outgrowth, InstiGRO samples were requested to conduct an evaluation.

Samples of InstiGRO CHO and InstiGRO CHO PLUS cell culture supplements were sent to the customer. While both are designed to enhance clonal outgrowth, the formulations differ slightly to cater for the different CHO cell lines and basal culture mediums used in the manufacture of protein therapeutics.

 

Method

7 CHO-S cell lines, each of which were expressing a different monoclonal antibody (mAb), were single cell seeded (via limiting dilution) into 384 well plates containing CD CHO medium. 128 wells were seeded with CD CHO medium supplemented with 5% InstiGRO CHO, amino acids, and L-Glutamine. A further 128 wells were seeded with CD CHO medium which was supplemented with 5% InstiGRO CHO PLUS, amino acids, and L-Glutamine. A final 128 wells were seeded with CD CHO medium with amino acids and L-Glutamine alone to provide a negative control. After 14 days, the number of colonies which emerged was totalled.

 

Results

The number of clones more than doubled when InstiGRO CHO or InstiGRO CHO PLUS was added to the cell culture medium.

Homing in on the different InstiGRO formulations, you can see that this customer’s cell lines preferred InstiGRO CHO PLUS to InstiGRO CHO. Remember, this is not always the case as the CHO cell line used and cell culture medium which InstiGRO is paired with will make a difference.
